Leishmania, the causative agent of leishmaniasis, has a unique organelle called a glycosome that compartmentalizes many vital metabolic pathways and its biogenesis and function depends on the glycosomal protein translocation system. Interaction between LdPEX14 and LdPEX5 on the glycosomal membrane is a crucial step for targeting proteins into this organelle. We used deletion mutagenesis to determine the minimal domain of the LdPEX14 N-terminal region that is critical for mediating the LdPEX5-LdPEX14 interaction.
